Background aimsMesenchymal stem/stromal cells (MSCs) are of interest for the treatment graft-versus-host disease, autoimmune diseases, osteoarthritis and neurological cardiovascular diseases. Increasing numbers clinical trials emphasize need standardized manufacturing these cells. However, many challenges related to diverse isolation expansion protocols differences in cell tissue sources exist. As a result, products used numerous vary greatly characteristics potency.MethodsThe authors have established culture platform using xeno- serum-free commercial media MSCs derived from umbilical cord (UC), bone marrow adipose-derived (AD) examined their functional characteristics.ResultsMSCs tested stably expanded vitro retained biomarker expression normal karyotype at early later passages after cryopreservation. were capable colony formation successfully differentiated into osteogenic, adipogenic chondrogenic lineages. Pilot UC-MSCs AD-MSCs scale revealed that met required quality standard therapeutic applications.ConclusionsThe authors’ data suggest conditions suitable large-scale enable comparative study different origins.
